How half-life is measured

Half-life is determined by sampling peptide concentration at defined intervals in a chosen matrix (plasma, serum, buffer, cell culture medium) and fitting the decay to a pharmacokinetic model. Values can differ by an order of magnitude between matrices and between species.

Why values vary

Reported half-lives depend on: enzymatic environment, species, route, dose, assay sensitivity, and whether the peptide is modified (e.g. PEGylated, lipidated, or DAC-bound). A peptide's half-life in plasma is not the same as its half-life in cell culture or tissue.
